摘要 β3肾上腺素能受体(ADRB3)在高浓度的儿茶酚胺作用下主要作用于脂肪组织调节脂肪分解和产热效应。研究采用PCR-SSCP和测序技术,对12个绵羊品种(系)共962只个体的ADRB3基因的部分序列进行了多态性分析。共检测出A~H等8种基因型,其中基因型B和C的频率较高,为优势基因型,基因型H的频率最低,且只在一个品种中发现。序列测定及比对结果共发现10个核苷酸变异位点,包括一处A碱基的插入。群体的有效等位基因数为2.240,等位基因丰度为6.515。各品种(系)的基因多样性值介于0.050-0.729之间,山西细毛羊的最小,小尾寒羊的最大。表明除山西细毛羊外,其它11个绵羊群体均具有较高的遗传多样性。 The beta-3 adrenergic receptor(ADRB3)is the main mediator of the lipolytic and thermogenic effects of high catecholamine concentrations.In this studyt,he polymorphism of ADRB3 gene in 962 individuals from 12 sheep populations were analysed by single strand conformation polymorphism technique based on PCR(PCR-SSCP)and sequencing.Eight genotypes named A,B,C,D,E,F,Ga,nd Hr,espectively,were detected.Allele B was the most common allele,with a frequency of 38.7 %f,ollowed by allele Ca,t 33.5 %.Allele H had the lowest frequency(0.1 %)and was found only in a single population.Ten polymorphic sites were detected by sequencing and alignmenti,ncluding a adenine insertion.The genetype diversity ranged from 0.050 in Shanxi finewool sheep to 0.729 in Small-tailed Han sheepi,ndicating abundant genetic diversity in the populations,except Shanxi finewool sheep.The effective number of alleles and allelic richness were 2.224 and 6.515 overall populationsr,espectively.
