血管紧张素Ⅱ1型受体基因A1166C在汉族人群中的分布及其与家族性高血压的相关性 被引量:1

The distribution of angiotensin Ⅱ type 1 receptor gene A1166C in Han nationality and the relationship between genetic polymorphism and Familial Primary Hypertension

摘要 目的 研究血管紧张素Ⅱ1型受体(AT1R)基因A1166C在正常汉族人群中的分布特征,及其与家族性原发性高血压的相关性.方法 选取健康成年人150例(正常对照组)和家族性原发性高血压患者178例(家族性高血压组),提取所有受试者外周血白细胞DNA,PCR-RFLP方法进行AT1R基因A1166C多态性检测.结果 正常汉族人群中AA、AC和CC基因型及该点A、C等位基因频率分布,在不同性别及年龄间比较均无显著性差异(P〉0.05);家族性高血压组基因型分布[AA 131(73.6%)、AC 41(23.03%)、CC 6(3.37%)],等位基因频率分布[A 303(85.11%)、C53(14.89%)],与正常对照组比较差异均有明显统计学意义(P〈0.05).结论 AT1R基因A1166C在成人汉族人群中的分布特征与性别、年龄无关;AT1R基因A1166C多态性与家族性原发性高血压明显相关,1166C携带者可能有较高患病风险. Objective To observe the distribution characteristics of angiotensin Ⅱ type 1 receptor (AT1R) gene A1166C in Han nationality and study the relationship of polymorphism and familial primary hypertension. Methods 178 patients with familial primary, hypertension and 150 healthy adults were included. DNA was drawn out by low permeate hemolysis and salting-out technique. The A1166C gene polymorphisms of AT1R were determined with the use of standard polymerase chain reaction methods and restriction fragment length polymorphism(PCR-RFLP). Results In normal control group,we observed that there were no differences between the various sub-groups divided by age or sex( P 〉0.05 ) ;In familial hypertension group,the genotype frequencies of A1166C were AA 131 (73.6%) ,AC 41 (23.03%) ,CC 6 (3.37%) ,the A1166 and C1166 allele frequencies were A 303(85.11% ) ,C 53( 14.89% ). There were notable statistic significances compared with the normal control group ( Х^2 = 7.75, Х^2 = 8.29, P 〈 0. 05 ). Conclusion The frequency distribution characteristic of AT1R gene Al166C in Han people is not related with gender and age; A significant association perhaps exists between the AT1R Al166C gene polymor- phisms and familial primary hypertension.
