摘要
采用相关序列扩增多态性(SRAP)技术分析了豌豆和玉米远缘杂交后代的真实性。结果表明,10对SRAP引物中,共扩增出105条重复性高的清晰条带,其中有101条为多态,多态比率(PPB)为96.19%;聚类结果显示,后代与母本间呈现出并列关系,表明其基因组确实发生了改变;em1-me2e、m3-me1和em8-me4三对引物在杂交后代中都扩增出双亲特异带,说明这2个杂交后代具有双亲的遗传物质,该后代是豌豆和玉米成功远缘杂交的真实杂种后代。
The truth of the hybrid progenies from wide cross of pea and maize was analyzed by using sequence-related amplified polymorphism(SRAP) markers.The results showed that among 10 pairs of primer,105 clear and reproducible bands were obtained,in which 101 bands were polymorphic,and the average polymorphic ratio was 96.19%.Clustering results showed that the hybrid progenies showed a parallel relationship with theirs female parent,which indicated that its genome was indeed changed;three pairs of primer(em1-me2,em3-me1 and em8-me4) all produced the specific markers of parents in the hybrid progenies,which indicated that the progenies contained some genetic material of both parents,and they were the true hybrid progenies from a success wide cross of pea and maize.
