摘要
目的研究天麻钩藤饮对帕金森病(Parkinson's disease,PD)模型大鼠多巴胺能神经元凋亡的影响。方法采用6-羟基多巴胺注射于脑右侧黑质造成单侧PD损毁模型,并将造模成功的大鼠随机分为模型组和天麻钩藤饮组(给药浓度为5.13g/ml),共给药14天。同时设立正常对照组、假手术组。应用TUNEL、透射电镜观察多巴胺能神经元凋亡的变化,免疫组化法观察黑质Bcl-2、Bax的表达。结果模型组均能检测到TUNEL染色阳性细胞核,且模型组凋亡细胞数明显多于正常对照组、假手术组(P<0.01)。天麻钩藤饮组也能检测到凋亡细胞核,但与模型组比较,凋亡细胞数较少(P<0.01)。与正常对照组及假手术组比较,模型组Bax和Bcl-2表达水平均显著升高(P<0.01),且以Bax蛋白升高为主;而与模型组比较,天麻钩藤饮组Bcl-2显著升高,Bax显著降低(P<0.01)。电镜结果显示,天麻钩藤饮组神经细胞病理变化比模型组明显为轻。结论天麻钩藤饮对PD模型大鼠多巴胺能神经元凋亡有明显抑制作用,其作用机制可能是通过抗氧化应激,升高Bcl-2,抑制Bax激活而实现的。
Objective To study the effect of Tianma Gouteng Yin (Decoction of Gastrodia and Uncaria) on apoptosis of dopaminergic neurons in Parkinson's disease (PD) model rats.Methods The PD models were established by injecting 6-hydroxy dopamine into the right substantia nigra of brain and treated with Tianma Gouteng Yin (5.13g/ml) for 14 days. At the same time the normal control group and pseudo-operation group were set up for comparison. The changes of dopaminergic nerons apoptosis were observed with the help of TUNEL and transmission electron microscopy and the expression of substantia nigra Bcl-2 and Bax was observed with immunohistochemical method.Results The positive TUNEL stained nucleus could be detected on the samples in the model group and the apoptosis cells were significantly more than those of the normal control group and pseudo-operation group (P〈0.01). In the Tianma Gouteng Yin group,there was also apoptotic nucleus found,but the apoptotic cells were much less than in the model group (P〈0.01). Compared with the normal control group and pseudo-operation group,the expression of Bax and Bcl-2,especially Bax,in the model group was significantly higher. Compared with the model group,the expression of Bcl-2 in the Tianma Gouteng Yin group was significantly higher,but that of Bax was significantly lower (P〈0.01). The electron microscopy examination showed that the pathological changes of neurons in Tianma Gouteng Yin group were significantly milder than those in the model group.Conclusion Tianma Gouteng Yin restrains significantly the apoptosis of dopaminergic neurons in rats with Parkinson's disease and its mechanism may be actualized by lifting the expression of Bcl-2 and restraining the activation of Bax through anti-oxidation stress.
