参麦注射液对肠缺血再灌注肺脂质过氧化损伤的影响 被引量:6

Effects of ShenMai Injection on Lung Lipid Peroxidation in Rat Model of Intestinal Ischemia/Reperfusion

摘要 目的:探讨参麦注射液对肠缺血/再灌注(I/R)肺脂质过氧化损伤的防护作用。方法:SD大鼠24只,随机均分为3组。对照组只分离出肠系膜上动脉;模型组、治疗组复制大鼠肠I/R损伤模型;治疗组于缺血前、再灌前、再灌后30min尾静脉注射参麦注射液0.9mL/100g体质量。观察各组外周血白细胞黏附聚集(LAA)、肺组织湿/干比(W/D)、肺系数以及血浆和肺组织超氧化物歧化酶(SOD)、丙二醛(MDA)、髓过氧化物酶(MPO)的变化及参麦注射液对上述指标的影响;并进行肺组织病理学检测。结果:(1)模型组LAA、肺组织W/D、肺系数均高于对照组(P<0.01),光镜下肺组织病理损害亦明显重于对照组;治疗组上述指标均明显小于模型组(P<0.01)。(2)模型组血浆和肺组织SOD活性低于对照组(P<0.01);MPO活性及MDA含量高于对照组(P<0.01),治疗组上述各检测指标均较模型组明显改善(P<0.01)。结论:参麦注射液对肠I/R后肺损伤具有保护作用,其机制可能与抑制中性粒细胞的聚集,对抗脂质过氧化损伤有关。 Objective: To explore the effects of shenmai injection(SM) on lung lipid peroxidation in rat model of intestinal ischemia/reperfusion (I/R). Methods: Twenty-four SD rats were randomly divided into three groups,sham group, I/R group and treatment group. The superior mesenteric artery was separated only in rats of sham group. Models of intestinal I/R injury were established in I/R group and treatment group. In treatment group, rats was injected SM (0.9 mL/100 g) through tail vein before ischemia, before reperfusion and 30 min after reperfusion, respectively. The values of leukocyte adhesiveness/aggregation (LAA) were detected in the peripheral blood, lung wet/dry weight ratio, lung coefficient, superoxide dismutase(SOD), malondialdehyde (MDA) and myeloperoxidase (MPO) in serum and lung tissue, and the effects of SM were observed as well. The changes of lung histopathology were examined by light microscope. Results:(1) The values of LAA in peripheral blood, lung wet/dry weight ratio and lung coefficient were significantly increased in I/R group than those in sham group (P 0.01). The results of light microscope examination showed that more serious damage of lung tissue in I/R group compared with those of sham group. The above-mentioned indexes were obviously lower in treatment group compared with those of I/R group (P 0.01). (2) The activity of SOD in serum and lung tissue was significantly lower in I/R group than that in sham group (P 0.01). The content of MDA and the activity of MPO in serum and lung tissue were significantly increased in I/R group than those in sham group (P 0.01). The above-mentioned indexes were obviously ameliorated in treatment group in comparison with I/R group (P 0.01). Conclusion: Shenmai injection can protect the lung injury induced by intestinal ischemia/reperfusion, which may be mediated by inhibiting lipid peroxidation and aggregation of neutrophils.
