摘要
目的应用二甲基亚硝胺(DMN)建立肝纤维化大鼠模型,研究以黄芪、丹参、三七为主的益气活血化瘀复方中药抗肝纤维化的作用机制。方法将Wistar大鼠随机分为正常组、模型组、中药干预组。采用DMN腹腔内注射的方法建立肝纤维化大鼠模型;根据病理学检查评价各组动物肝纤维化程度;应用RT-PCR检测各组肝组织中Ⅰ、Ⅲ型胶原和TGF-β1mRNA水平;应用免疫组化检测各组肝组织Ⅰ、Ⅲ型胶原和TGF-β1的表达。结果中药干预组大鼠肝纤维化程度、肝组织中Ⅰ型与Ⅲ型胶原和TGF-β1mRNA水平,以及肝组织中Ⅰ型与Ⅲ型胶原和TGF-β1的阳性表达均分别较模型组显著减轻与下降。结论益气活血化瘀复方中药能明显减轻肝组织病理改变和肝纤维化程度,抑制Ⅰ、Ⅲ型胶原和TGF-β1表达是其抗肝纤维化的部分机制。
Objective To investigate the anti-fibrotic effects of Chinese herbs compound of Nourishing Qi and Activating circulation on rats with liver fibrosis induced by dimethylnitrosamine. Methods Wistar rats were randomly divided into normal control group,model group,Chinese herbs compound intervened group. The animal models of liver fibrosis were induced by intraperitoneally injecting with dimethylnitrosamine (DMN ). Liver fibrosis were evaluated on the liver tissue sections which were performed with HE and Reticulation’s stain. Semi-quantitive RT-PCR was performed to determine mRNA levels of collagen type I and III and TGFβ1 in liver tissues. Immunohistochemical staining was performed to detect the expression of collagen type I and III and TGFβ1 in liver tissues. Results Compared with model group,the degree of liver fibrosis in Chinese herbs compound intervened group was obviously lowered. In intervened group,the mRNA levels of collagen type I and III,and TGFβ1 in liver tissue were all obviously lower than those in the model group respectively. The positive expressions of collagen type I and III and TGFβ1 in liver tissues of intervened group were all much weaker than those in the model groups respectively. Conclusion Chinese herbs compound of Nourishing Qi and Activating Blood Circulation has the positively preventive effects on the experimental liver fibrosis induced by DMN in rats,inhibiting the expressions of collagen type I and III and TGFβ1 in liver tissues is one of the anti-fibrosis effects of the Chinese herbs compound.
