摘要
以番茄“矮黄”、“鲜丰”品种的下胚轴为外植体进行离体培养,发现MS培养基上附加不同浓度的生长素(IAA)和细胞分裂素(BA),对愈伤组织的形成影响不大,但对不定芽的分化有较大影响,且品种间存在明显差异:品种“矮黄”,在MS+BA1.0~2.5mg·L-1+IAA0.2~0.5mg·L-1培养基上,诱导率都达50%以上,其中以MS+BA2.5mg·L-1+IAA0.2mg·L-1效果最佳,诱导率高达81.3%;品种“鲜丰”,在MS+BA1.0~2.0mg·L-1+IAA0.2mg·L-1培养基上,诱导率达50%以上,其中BA2.0mg·L-1时,诱导率最高,达76.3%。在同样培养基上继代培养,待芽长1~2cm时转到生根培养基上,可在短期内长出根系,形成小植株。
In this study, tomato hypocotyls cut from varieties Aihuang and Xianfeng were used as explants for culture on MS media supplemented with BA and IAA at different levels. The results showed that the effects of BA and IAA levels in media were less signifi-cant on callus formation, but more significant on adventitious shoot differentiation; there were some differences between varieties Aihuang and Xianfeng. For Aihuang,the shoot in-duction rate was over 5O% on MS media supplemented with 1. O^2. 5 mg BA/L+O. 2~O. 5mg IAA/L,and was the highest (81. 3% ) on MS media supplemented with 2. 5 mg BA/L+ O. 2 mg IAA/L. For Xianfeng,the shoot induction rate was over 5O% on MS media supple-mented with 1. O^2. O mg BA/L+O. 2 mg IAA/L,and was the highest (76. 3 % ) on MS me-dia supplemented with 2. O mg BA/L+O. 2 mg IAA/L. The adventitious shoots were subcul-tured on the same media and transferred onto rooting media (1/2 MS media) when growing to l^2 cm in length,and then produced roots and formed plantlets in a short time.
