摘要
目的:建立芬太尼在染毒家兔体内的毒物代谢动力学动物模型,研究芬太尼在染毒家兔体内的毒物代谢动力学的特征。方法:健康家兔6只,经耳缘静脉匀速注入芬太尼注射液2 mL(0.1 mg),给药后观察家兔的中毒症状,分别于给药后1,3,5,10,15,20,30,40,50,60 min,经颈总动脉取血1 mL,10%氢氧化钠调pH=10,乙醚萃取,气相色谱/质谱(GC/MS)定性、GC定量检测其中芬太尼。结果:中毒剂量芬太尼在家兔体内的毒物代谢动力学符合静脉给药一级动力学二室开放模型,动力学方程为C(T)=45.65e-0.24t+0.67e-0.003t,动力学参数:t1/2α=2.84 min,t1/2β=20.37min,K10=0.11,K21=0.07,K12=0.12,α=0.24,β=0.003,CL=128.66 L/min,AUC=388.60。结论:中毒剂量芬太尼在家兔体内毒物代谢动力学符合静脉给药一级动力学二室开放模型。依据所得动力学方程及所检测的浓度可以推算出给药后所经历的时间,此特征可为芬太尼麻醉意外中毒(死)案件的法医学鉴定提供一定的实验依据。
Objective:To establish a model of toxicokinetics of fentanyl in acute poisoned rabbits,and study the regulation of toxieokinetics of fentanyl in rabbits. Methods :Six rabbits were given fentanyl through vein of ear with a dose 2 mL(0.1 mg). The symptoms were observed and the 1 mL blood was collected at 1,3,5,10,15,20,30,40,50,60 min through common carotid artery after administration. The specimen was alkalified to pH 10,and then extracted with ether. The extraction was analyzed qualitatively and quantitatively by GC/MS and GC respectively. Results :The toxicokinetics of fentanyt in rabbits conform to single compartment open model with a first order processes by vein ad- ministration. The equation of dynamics is C(T)=45.65e^-0.24t+0.67e^-0.003t,动力学参数:t1/2α=2.84 min,t1/2β=20.37min,K10=0.11,K21=0.07,K12=0.12,α=0.24,β=0.003,CL=128.66 L/min,AUC=388.60. Conclusion :The toxicokinetics of fentanyl in rabbits with a poison dose conform to single compartment open model with a first order processes by vein administration. The time of administration can be estimated by the equation of dynamics and the concentration. This characteristic can provide a certain experimental basis and a scientific evidence for forensic identification of the poison caused by fentanyl.
