秦公散对LPS活化的内皮细胞表达黏附分子和分泌一氧化氮的影响

摘要 为阐明秦公散的抗炎作用机制,本试验采用流式细胞术检测了LPS刺激的内皮细胞黏附分子E-选择素和ICAM-1的表达,用一氧化氮(NO)测定试剂盒检测LPS刺激的内皮细胞培养上清液中NO的含量。结果表明,LPS刺激的内皮细胞可显著提高E-选择素和ICAM-1的表达(P<0.01),显著提高培养上清液中NO的含量(P<0.05);用秦公散及其含药血清处理后则明显抑制LPS诱导的E-选择素、ICAM-1的表达和NO分泌(P<0.05)。表明秦公散可通过抑制黏附分子的表达及NO分泌量发挥抗炎作用。 To state the Anti-inflammatory effects mechanism of Qingongsan,the flow cytometry was adopted to detect the expression of E-selectin and ICAM-1 in the endothelial cell activated by LPS and the NO determining kit was adopted to determine the concentration of NO in cultivation supernatant.Results: the human umbilical vein endothelial cells were cultured with LPS together can remarkably increase the expression of E-selectin and ICAM-1(P0.01),treated by crude extract of Qingongsan and the serum containing drugs can inhibit the expression of E-selectin,ICAM-1 and increase secretary of NO induced by LPS(P0.05).Qingongsan can educe its anti-inflammation function though inhabiting the expression of adhesion molecule and excessive secretion of NO.
