摘要
目的为肝癌的治疗提供新思路。方法将对数生长期肝癌Hep1细胞随机分为6组,空白对照组仅加入培养液,实验对照组加入等量溶媒二甲基亚砜(DMSO),姜黄素1~4组分别加入姜黄素5、10、15、20μmol/L。各组均置于常氧环境中培养24 h。RT-PCR法检测HIF-1αmRNA表达,Western blot法检测HIF-1α蛋白表达。结果姜黄素2~4组HIF-1αmRNA表达均明显高于空白对照组(P均<0.05)。姜黄素1~4组HIF-1α蛋白表达均明显高于空白对照组(P均<0.01)。结论常氧条件下,姜黄素呈浓度依赖性方式上调肝癌Hep1细胞HIF-1α的表达。
Objective To provide new ideas for the treatment of hepatocellular carcinoma.Methods Hepatocellular carcinoma Hep1 cells in logarithmic growth were randomly divided into 6 groups,the blank control group was treated with nutrient solution,the experimental control group was treated with equal amount of dimethyl sulphoxide,curcumin 1-4 groups were treated with curcumin of 5,10,15,20 μmol / L.All the groups were cultured under normal oxygen conditions for 24 hours.HIF-1α mRNA expression was measured by RT-PCR,and HIF-1α protein expression was measured by Western blot.Results HIF-1α mRNA expression in curcumin 2-4 groups were all higher than that in blank control group(all P 0.05).HIF-1α protein expression in curcumin 1-4 groups were all higher than that in blank control group(all P0.01).Conclusion Under normal oxygen conditions,curcumin can increase HIF-1α expression in hepatocellular carcinoma Hep1 cell in concentration-dependent manner.
