摘要
目的 观察新生鼠缺氧缺血性脑损伤(HIBD) 后脑组织中神经元特异性烯醇化酶(NSE) 、胶质纤维酸性蛋白(GFAP) 、S100 蛋白(S100)m RNA表达的变化。 方法 采用7 日龄SD大鼠缺氧缺血性脑损伤模型,用逆转录聚合酶链反应(RTPCR) 方法,动态观察脑组织中NSE、GFAP、S100 蛋白mRNA 水平变化。 结果 NSE m RNA 转录高峰出现在缺氧缺血后24 小时;GFAP、S100 蛋白m RNA转录高峰出现在缺氧缺血后48 小时,以后都随时间变化逐渐下降。 结论 在缺氧缺血性脑损伤后有NSE、GFAP、S100 蛋白m RNA 表达增加。提示HIBD 时在血和脑脊液中NSE、GFAP、S100 水平的升高,除从损伤细胞漏出外。
Objective To observe the expression of mRNA for neuron specific enolase (NSE), glial fibrillary acidic protein(GFAP)and S 100 protein (S 100) in cerebral tissue after hypoxic ischemic brain damage(HIBD) in newborn rat. Methods The dynamic changes of expression of mRNA for NSE、GFAP and S 100 protein in the cerebral tissue were studied by using 7d old SD rat hypoxic ischemic brain damage model and RT PCR technique. Results The peak expression of mRNA for NES and for GFAP and S 100 protein occurred 24 and 48 hours after HIBD respectively, and their expression decreased gradually. Conclusion The expression of mRNA for NSE、GFAP and S 100 protein increased significantly after HIBD. It suggests that the increased level of NSE、GFAP and S 100 protein in serum and cerebrospinal fluid (C.S.F) maybe reflect the increase of transcription and transnational except for the leakage of damaged cell.
