摘要
目的观察乌头类生物碱(双酯型生物碱与单酯型生物碱)对慢性心力衰竭(CHF)模型大鼠心肌组织中超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量的影响。方法采用腹主动脉缩窄法,制备慢性心力衰竭大鼠模型,8周后,用16导生理记录仪采集并分析心率(HR)、左心室舒张末期压(LVEDP)、左心室收缩压峰值(LVSP)、左心室内压最大上升/下降速率(±dp/dtmax)等心功能指标。心力衰竭模型成功后,给药组灌胃给予生物碱2周,测定心肌组织SOD活力、MDA含量。结果造模组SOD值明显低于假手术组,MDA值显著高于假手术组(P<0.05)。各给药组SOD值降低明显,MDA值升高显著(P<0.05)。结论乌头类生物碱能明显降低大鼠心肌匀浆中SOD的活性,同时增加氧化产物MDA的含量,提示乌头类生物碱对心力衰竭大鼠的心肌毒性机制与心肌氧化损伤有关。
Objective To observe the effects of diester-diterpenoid alkaloids and monester-diterpenoid alkaloids on myocardial SOD activity and MDA content in congestive heart failure rats.Methods The rat model of chronic heart failure was established by Abdominal Aortic Coarctation,and after eight weeks,the cardiac function indices,including heart rate（HR）,left ventricular end-diastolic pressure（LVEDP）,left ventricular systolic pressure（LVSP）,the maximal/ minimal derivative of left ventricular pressure（±dp/dtmax）were analyzed with BioPac System.After the model of heart failure succeeded,the rats were intragastric administered alkaloids for two weeks and the activity of SOD and the content of MDA in myocardial tissue were measured.Results The activity of SOD in model group were significantly lower than that of sham-operated group（P0.05）,the content of MDA in model group was significantly higher than that of sham-operation group（P0.05）,aconitum alkaloids could significantly reduce the activity of SOD in myocardial homogenate,the content of MDA.Conclusion The effects of aconitum alkaloids on cardiac toxicity is related to the oxidative damage of myocardium.
