星点设计-效应面法及正交实验设计优化淫羊藿提取工艺的比较研究 被引量:18

摘要 目的采用正交设计与星点设计-效应面法两种实验设计方法,优选淫羊藿提取工艺,并对设计方法进行比较。方法以淫羊藿苷提取量为因变量,乙醇浓度、回流时间和溶剂(倍)量为自变量,优选提取工艺。结果正交实验设计确定最佳工艺是12倍量60%乙醇回流提取两次,3 h/次;星点设计效应面优化法确定最佳工艺是12倍量50%乙醇回流提取两次,160 min/次。结论星点设计-效应面法在该提取工艺研究中优于正交实验设计法,为其应用于中药提取工艺优化的可行性提供了依据。 Objective To optimize the process of extacting effective constituents from Epimedii Herba by orthogonal experimental design and central composite design-response surface method.Methods Independent variables were concentration of ethanol,extraction time,times of reflux and solvent fold.Dependent variable was extraction rate of icariin in Epimedii Herba.Linear or nonlinear mathematic models were used to estimate the relationship between independent and dependent variables.Results The result of the orthogonal experimental design was 60% ethanol,180 minutes for reflux,12 fold of solvent and 2 times for extraction.The other was 50% ethanol,160 minutes for reflux,12 fold of solvent and 2 times for extration.Conclusion The Central Composite Design-Response Surface Method is highly predictive in experimental design.
