雄激素对前列腺中miR-204表达调控的研究

摘要 目的:探讨雄激素对前列腺癌细胞和组织中miR-204表达的影响。方法:采用荧光定量PCR方法检测前列腺癌细胞株(LNCaP、22RV1)和正常前列腺间质细胞(WPMY-1)在雄激素类似物R1881刺激下以及去势后不同时间的小鼠前列腺组织中miR-204表达量的变化。结果:在前列腺癌细胞LNCaP、22RV1中hsa-miR-204的表达量在雄激素刺激后下降了近2倍,而在去势小鼠的前列腺组织中,mmu-miR-204表达量增加了1.8倍。结论:miR-204受雄激素的负调控。 Objective: To explore whether the expression of miR-204 was regulated by androgen in prostate cancer cells and tissue.Methods: miR-204 expression was detected among prostate cancer cells (LNCaP,22RV1) and normal stromal cell line WPMY-1 with/without 2 nmol/L androgen analog R1881 respectively,also in prostate tissue of castrated mouse using quantitative Real-time RT-PCR,data were analyzed using the 2-△△Ct method.All experiments were carried out in triplicate.Results: The expression of miR-204 showed nearly 2-fold decrease after androgen treatment in LNCaP and 22RV1 cells,respectively.However,there was no change in WPMY-1 cells.The mmu-miR-204 showed about 1.8-fold increases in prostate tissue of castrated mouse.Conclusion: The miR-204 was an androgen-responsive miRNA and downregulated by androgen analog R1881.
