摘要
目的:采用HPLC测定利咽消颗粒和传统汤剂中甘草酸、甘草苷、龙胆苦苷的含量,提供利咽消颗粒质量控制的方法。方法:选用Shim-pack VP-ODS柱,以甲醇—0.2%醋酸铵(67∶33)为流动相,在250nm波长测定甘草酸;选用Diamonsil C18柱,流动相分别以乙腈—0.5%冰醋酸(20∶80),在276nm波长测定甘草苷;以甲醇—水(40∶60),在254nm波长测定龙胆苦苷。结果:方中甘草酸、甘草苷、龙胆苦苷在传统汤剂和颗粒剂中的含量分别为0.2878、0.1548、0.0187 mg/g和0.6253、0.1818、0.022 mg/g,颗粒剂中平均含量均大于传统汤剂中平均含量;3个成分组间无重复双因素方差分析表明P<0.05。结论:颗粒剂中3种成分的含量高于传统汤剂中的含量,3种成分峰都能有效分离,其测定方法简便、准确、重复性良好,方中其它成分对测定无干扰,可以作为质量控制的方法。
Objective:Glycyrrhizin、glycyrrhizic acid、gentiopicrin in Liyanxiao particle and traditional decoction can be determined by HPLC,to Provide the quality control for Liyanxiao particle.Methods:the Shim-pack VP-ODS col-umn was used for glycyrrhizic acid,the mobile phase was methanol-0.2% Ammonium acetate(67 ∶ 33),detection wavelength at 250nm;the Diamonsil C18 column was used for glycyrrhizin and gentiopicrin,the mobile phase was acetoni-trile-0.5% glacial acetic acid(20 ∶ 80),detection wavelength at 276nm;And the mobile phase was methanol-water(40 ∶ 60),gentiopicrin can be determined at 254nm.Results:The respective contents of glycyrrhizic acid,glycyrrhizin and gentiopicrin is 0.2878、0.1548、0.0187mg/g and 0.6253、0.1818、0.022mg/g in traditional decoction and particl,the average contents in particl were more than those in the traditional decoction,two-factor analysis of variance indicates three ingredients without duplicate interblock P 〈 0.05.Conclusion: The contents of three ingredients in particl were more than those in the traditional decoction,the peaks of three ingredients are segregated effectively,and the method is simple,conveient,exact and has good reproducibility,the other ingredients do not have interferences for the determination,and can be used in the quality control of Liyanxiao particle.
