摘要 目的:研究大黄素对体外培养的人脐静脉血管内皮细胞(HUVEC)的增殖和凋亡的影响。方法:体外培养HUVEC,利用MTT法评价不同浓度的大黄素(10、20、40、80μmol·L-1)对HUVEC增殖的影响;观察其对HUVEC细胞形态的影响;利用DNA梯状条带观察不同浓度大黄素对HUVEC凋亡的影响。结果:不同浓度的大黄素作用于HUVEC24、48、72h后,可明显抑制细胞的增殖,并能够导致细胞发生皱缩和变圆;不同浓度大黄素作用HUVEC细胞72h以后,细胞出现明显的DNA梯状条带。结论:大黄素对体外培养的HUVEC的增殖具有抑制作用,而这种作用可能是通过抑制细胞增殖、促进细胞凋亡而产生的。 OBJECTIVE:To study the effect of emodin on the proliferation and apoptosis of human umbilical vein endothelial cells(HUVEC).METHODS:The HUVEC were cultured with different concentration of emodin(10,20,40,80μmo·lL -1)in vitro.The effect of emodin on the proliferation of HUVEC was evaluated by MTT assay.The effect of emodin on the morphology of HUVEC was observed.The DNA ladder was used to observe the effect of emodin at different concentrations on the apoptosis of HUVEC.RESULTS:The different concentration of emodin inhibited the proliferation of HUVEC significantly and induced the morphological change of cells at 24 h,48 h and 72 h after treatment.Obvious DNA ladder was observed at 72 h after treatment.CONCLUSION:Emodin can inhibit the proliferation of HUVEC,which promotes the apoptosis of HUVEC.
