Simultaneous determination of sophoricoside and rutin content in Fructus Sophorae by high performance liquid chromatography

摘要 目的建立同时测定槐角中槐角苷和芦丁含量的高效液相色谱法(HPLC)。方法采用HPLC法对采集到的10批槐角药材中的槐角苷与芦丁的含量进行测定,Agilent ZORBAX-C18柱(4.6mm×250.0mm,5μm),流动相为:甲醇-乙腈-0.007mol·L-1磷酸溶液(体积比122068),流速为1.0mL·min-1,柱温:30℃,检测波长为257nm。结槐角苷在2.40~48.00mg·L-1、芦丁在1.15~23.00mg·L-1范围内线性关系良好;精密度试验槐角苷和芦丁的相对标准偏差(RSD)分别为1.05%和0.87%;供试品在室温放置24h后槐角苷和芦丁含量的RSD分别为1.78%和1.34%;槐角苷和芦丁供试品溶液的平均含量分别为6.71%和3.52%,RSD分别为1.45%和1.37%;槐角苷和芦丁供试品溶液回收率分别为100.2%和100.4%,RSD分别为1.63%和1.45%。结论本高效液相色谱法简便、准确,可同时测定槐角中槐角苷和芦丁的含量。 Objective To develop an high performance liquid chromatography ( HPLC) method for the simultaneous determination of sophoricoside and rutin content in Fructus Sophorae. Methods Sophoricoside and rutin contents in Fructus Sophorae of ten batch fructus medical material collected were simultaneously determined. The Agilent ZORBAX-C18( 4. 6 mm ??250. 0 mm,5 ??) column with Methanol-Acetonitrile -0. 007 mol?? L -1 NaH2 PO4 ( V:V:V = 12:20:68) as the mobile phase,the flow rate was 1. 0 mL??min -1 ,the column temperature was 30 ?校nd the detection wavelength was 257 nm. Results The linear correlation between sophoricoside and rutin worked well in the range of were 2. 40 -48. 00 mg??L -1( sophoricoside) and 1. 15 -23. 00 mg?? L -1 ( rutin). Relative standard deviation ( RSD) of sophoricoside and rutin in precision testing were 1. 05% and 0. 87% respectively. RSD were 1. 78% and 1. 34% respectively after 24 hours at ambient temperature. The average content of sophoricoside and rutin solution were 6. 71% and 3. 52% ,RSD were 1. 45% and 1. 37% respectively; The recovery rates were 100. 2% and 100. 4% ; RSD were 1. 63% and 1. 45% respectively. Conclusion The HPLC method is simple and accurate and it can be used to simultaneously determine sophoricoside and rutin content in Fructus Sophorae.
