摘要
目的探讨穿孔素(PFP)介导的细胞毒效应在病毒性心肌炎(VMC)发病中的作用。方法40只小鼠等分为CVB3感染组及非感染组;所有小鼠于感染后第10天处死并取其心脏;采用免疫组化技术检测心肌浸润细胞中PFP的表达,原位杂交方法检测心肌中CVB3RNA;半定量分析PFP抗原及CVB3RNA水平,并分析PFP抗原水平与CVB3复制及心肌病变程度的关系。结果PFP抗原阳性信号指数与CVB3RNA杂交阳性信号指数呈显著负相关(r=-0.82,P<0.05),与心肌病变积分呈显著正相关(r=0.91,P<0.05)。结论PFP介导的细胞毒效应在VMC发病中可能具有抗病毒及致心肌损害双重作用。
Purpose To clarify effect of perforinmediated
cytotoxicity in the development of murine acute myocarditis. Methods Forty mice were equally
divided into experimental group(CVB3 infected mice) and control group (non infected mice).The
mice were killed and their hearts were removed 10 days after infection. The expression of
perforin in infiltrated cells of myocardium was determined by immunohistochemistry. CVB3
RNA in myocardium was detected by in situ hybridization. In addition,the levels of perforin
antigen and CVB3 RNA were semiquantitatively analyzed, and the relationship of the perforin
expression level with the degrees of CVB3 replication and myocardial lesions was investigated.
Results The fraction of perforin antigen positive signal show significantly correlation with that
of CVB3 RNA negative signal( r=-0.82,P<0.05) and demonstrated significantly positive
correlation with myocardial histopathological score ( r=0.91,P<0.05 ). Conclusions Perforin
mediated cytotoxicity might play an important role in limiting virus replication and promoting
cardiac damages.
