摘要
目的观察连接蛋白43(Cx43)在乳腺癌组织及癌前病变组织中的表达,探讨Cx43在乳腺癌发生过程中的作用机理。方法应用S-P免疫组织化学方法,检测Cx43在20例乳腺癌组织、20例乳腺导管上皮内瘤变组织、10例良性乳腺病变组织和10例癌旁正常乳腺组织中的表达。应用RT-PCR和Western blot法,检测Cx43 mRNA及Cx43蛋白在四组乳腺组织中的表达。结果乳腺癌组织与乳腺导管上皮内瘤变组织中Cx43蛋白表达减少,且Cx43蛋白在乳腺癌组织中的表达明显低于乳腺导管上皮内瘤变组织中的表达(P<0.01)。RT-PCR显示乳腺导管上皮内瘤变组织中Cx43 mRNA的表达量减少,而乳腺癌组织中Cx43 mRNA的表达量明显减少(P<0.01)。结论 Cx43蛋白低表达及其构成的缝隙连接细胞间通讯异常,与基因转录受到抑制有关,从而可能导致乳腺上皮细胞分化异常和过度增生,促进了乳腺癌的发生。
Objective To investigate the expressions of Cx43 of breast cancer and ductal intraepithelial neoplasia,and to probe the regularity and significance of Cx43 of breast cancer.Methods We collected the 20 breast cancer tissues,20 ductal intraepithelial neoplasia,10 benign lesions of breast and 10 normal tissues,then S-P method was used to detect Cx43,RT-PCR and Western blott method were used to observe the difference of Cx43 mRNA and Cx43 protein.Results A little of position could find at the touch of cells and major of them in endochylema in ductal intraepithelial neoplasia(DIN).It was expressed mainly in endochylema and obviously reduced of position in carcinoma of breast tissues(P 0.01).RT-PCR showed that Cx43 mRNA was lower in DIN and obviously reduced in breast cancer than other two groups(P 0.01).Conclusion The expressions of Cx43 reduced in breast cancer,maybe because of inhibition of transcription,that may involve the prosoplasia and hyperplasia excessively of epithelial cells and development of breast cancer.
