STR对幼鲤生长和肝脏IGF-Ⅰ mRNA表达的影响 被引量:2

Effects of STR Injection on Growth and Hepatic IGF ⅠmRNA Expression in Common Carp

摘要 通过以体质量60~70g的幼年鲤鱼(Cyprinuscarpio),每kg体质量以200mg的剂量体腔注射链脲佐菌素(streptozotocin,STR)或载体溶液200mg,观察鲤鱼生长、血糖、血清生长激素(GH)水平和肝组织胰岛素样生长因子Ⅰ(IGFⅠ)mRNA水平的变化.注射STR的鱼生长速率比对照组显著减慢.在第15天和第30天实验组鲤鱼血糖水平以及血清GH水平皆无明显变化.在注射STR后的第15天肝组织IGFⅠmRNA的表达水平比对照组明显下降,第30天两组鱼肝组织IGFⅠmRNA含量则无明显差异.注射STR后鲤鱼出现胰岛遭受破坏的症状.实验结果提示STR可能通过破坏胰岛B细胞导致胰岛素(Ins)分泌下降,而后者则可能通过GH受体水平或GH受体以后水平的某种机制导致鲤鱼肝组织IGFⅠmRNA表达的下降。 Juvenile common carp, about 60~70 g, were injected intraperitoneally either with 200 mg/kg body wt STR, an agent known to induce damage of pancreatic B cells in mammals, or with the vehicle in order to study the effect of STR on the growth and hepatic IGF ⅠmRNA expression in common carp The control fish grew quickly throughout the experiment, while the growth of STR injected fish was retarded There was no difference between the two groups of fish in plasma levels of either glucose or growth hormone In corresponding with the fish growth state, on the day 15 post STR injection, the hepatic IGF ⅠmRNA level declined significantly On the day 30, the hepatic IGF ⅠmRNA level of STR injected fish was somehow restored These results imply STR injection may induce hypoinsulinemia by causing pancreatic B cells destruction to some extent and then lead to diminished hepatic IGF Ⅰ production, which eventually lead to the growth retardation of STR treated common carp
