摘要
从病毒感染后期的斜纹夜蛾幼虫脂肪体中提取mRNA,反转录合成cDNA探针,并使之与SlNPV基因组的酶切片段杂交,将SlNPVp10基因定位于BamHⅠ30千碱基片段上.SlNPVp10是目前发现的最长p10基因;基因的启动子内含有2个ATTGTA基序,是目前所发现的p10基因中唯一含有2个A/TTTGTA基序者.SlNPVP10蛋白含有10个七肽重复单元,可形成1个相对较长的卷曲螺旋.SlNPVp10所在的ORF552p10ORF945基因簇与SpliNPV的ORF552p10ORF945基因簇各基因座位、方向及对应的氨基酸序列十分相似。
The p 10 gene was localized within the 3 0 kb Bam HⅠ fragment of the Sl NPV genome by Southern hybridization. With a coding sequence of 318 nucleotides, corresponding to a protein of 105 amino acids, the p 10 gene of Sl NPV is the longest p 10 gene identified so far in NPVs. The Sl NPV p 10 gene is also distinct in having two A/TTTGTA motifs in the promoter region. With 10 heptad repeats, the Sl NPV P10 protein is probably forming a relatively large coiled coil structure. The organization of Sl NPV ORF552 p 10 gene ORF945 cluster is colinear with that of Spli NPV and their corresponding amino acid sequences of each ORF share high similarity. This indicates that Sl NPV and Spli NPV belongs to a homologous baculovirus group.
