白芍提取物对慢性应激抑郁模型大鼠行为学及大脑皮质单胺类神经递质的影响 被引量:45

Effects of extract of White Peony Root on the behavior and cerebral cortex monoamine neurotransmitters in depressive model of chronic unpredictable mild stress rats

摘要 目的:探讨白芍提取物的抗抑郁作用及可能机制。方法:以孤养加慢性轻度不可预见性应激方法建立大鼠抑郁症模型,用糖水消耗、敞箱行为及跳台行为测定进行行为学评分,并用高效液相-电化学方法检测其脑内单胺类神经递质的含量,观察模型大鼠给药前后的变化。结果:模型组的大鼠体质量增长缓慢,蔗糖水消耗量明显下降,水平活动和垂直活动均显著下降,跳台错误次数增加,脑内的去甲肾上腺素、5-羟色胺含量降低(P<0.05)。白芍提取物70mg.kg-1.d-1、35mg.kg-1.d-1剂量能显著改善模型大鼠的行为学变化,增加大鼠脑内去甲肾上腺素、5-羟色胺含量(P<0.05)。结论:白芍提取物具有抗抑郁作用,对中枢单胺类神经递质的调节作用是其作用机制之一。 Objective: To study the anti-depression mechanism of extract of Paeonia sterniana Fletcher in Journ. Methods: Rat models were established by separation and chronic unpredictable mild stress (CUMS), sucrose consumption test, open-field test and step-down passive avoidance test were used to observe the behavior in model rats. HPLC-ECD was used to analyze the levels of monoamine neurotransmitters. Results: Compared with normal group, the weight, the frequency of crossing and rearing, errors and the consumption of sucrosum water in depressive disorder rats decreased significantly (P0.05), The contents of 5-HT and NA reduced (P0.01). The extract of White Peony Root 70mg·kg -1 ·d -1 , 35mg·kg -1 ·d -1 could correct behavioral changes and increase the contents of 5-HT and NA (P0.05). Conclusion: The effects of anti-depression of the extract of White Peony Root may be related to the regulation of monoamine neurotransmitters in brain.
