The human immunodeficiency virus type 2 (HIV-2) gp36 DNA fragment was
amplified by polymerase chain reaction from the HIV-2 ROD 2 cDNA template using a pair of the
designed primers containing Nde Ⅰ and Xho Ⅰ sites, then was ligated with pGEM-T vector by
T4 DNA ligase to construct the plasmid pGEMT-36. The gp36 fragment cutted with NdeⅠ and
XhoⅠwas inserted into the expression plasmid pExSecⅠincluding these two endonuclease
sites for constructing the recombinant plasmid pExSec Ⅰ-36. It was confirmed by PCR,
restrcition enzyme and sequence analysis. The recombinant protein was expressed in E. coli
, rgp36 was induced by IPTG to produce the recombinant protein, its molecular weight and
antigenicity were confirmed by the analysis of SDS-PAGE and Western blot.
