聚腺苷二磷酸核糖聚合酶-1对热休克蛋白60的抑制作用

摘要 目的观察聚腺苷二磷酸核糖聚合酶-1(PARP-1)对早产儿脑室周围白质软化(PVL)小鼠模型及体外培养的少突胶质前体细胞中的热休克蛋白60(HSP60)表达的调控作用。方法建立未成熟小鼠野生型(Wild-type)和PARP-1基因缺失型小鼠脑白质损伤模型,术后取脑,冰冻切片,采用免疫荧光标记法对切片内HSP60进行染色,并在荧光显微镜下观察HSP60的表达。体外培养的少突胶质前体细胞经PARP-1的抑制剂3,4-dihydro-5-[4-(1-piperidinyl)butoxyl]-1(2H)-isoquinoline(DPQ)或激活剂Etopside预处理,再经IFN-r激活后,应用Western-blot方法检测细胞内HSP60的表达情况。结果在PARP-1基因缺失型小鼠脑白质损伤模型脑中,HSP60的表达量明显高于Wild-type组(P<0.05)。PARP-1抑制剂DPQ能够明显增强HSP60的表达,而PARP-1激活剂Etopside可显著抑制HSP60的表达。结论 PARP-1对HSP60的基因表达具有负调控作用。 Objective To observe the inhibitory effect of PARP-1 on the expression of HSP60 in PVL animal model and cultured OPC.Methods PVL animal model in wild-type or PARP-1 knockout mice were made and HSP60 was stained by anti-HSP60 antibody and observed under the fluorescence microscope.OPC were isolated and cultured,then pretreated with DPQ or Etopside before IFN-r was added.Cells were lysed and collected,HSP60 expression was checked by Western-blot.Results The expression of HSP 60 was significantly increased in PARP-1-/-compared with wild type PVL animal model.When treated with PARP-1 inhibitor DPQ or activator Etopside,HSP60 expression obviously increased or decreased correspondingly.Conclusion The expression of HSP60 can be negatively modulated by PARP-1.
