寡核苷酸CpG-ODN增强HBV疫苗接种的细胞免疫反应

摘要 目的研究寡核苷酸CpG-ODN对小鼠接种基因重组乙型肝炎病毒(HBV)疫苗产生的细胞免疫的影响。方法 BALB/c(H-2d)小鼠腹腔分别接种基因重组HBV疫苗(rHBs)或50μg CpG-ODN+rHBs,rHBs分为0.65、1.25、2.50、5.00μg剂量组。4周后分离小鼠脾淋巴细胞,体外分别用rHBs刺激,3H掺入实验检测特异性淋巴细胞增殖反应的液闪计数值(cpm)。结果 (1)接种0.65、1.25、2.50、5.00μg rHBs的小鼠脾淋巴细胞,体外用rHBs刺激,cpm值(x±s)分别为3 830.24±1 496.12、2 736.19±1 238.06、4 100.37±1 723.74、1 246.18±1 088.88,均高于体外未用rHBs刺激的对照组cpm值:1 607.00±182.6、1 677.5±358.32、255.5±227.3、753.8±206.8,差异有统计学意义(P<0.05)。(2)接种0.65μg rHBs+50.00μg CpG-ODN、1.25μg rHBs+50.00μg CpG-ODN、2.50μg rHBs+50.00μg CpG-ODN、5.00μg rHBs+50.00μg CpG-ODN的小鼠cpm值分别为6 863.87±1 243.24、6 582.48±1 132.48、7 332.43±938.285、184.24±947.45,均高于体外未用rHBs刺激的对照组cpm值:2 307.00±182.62、577.5±358.32、655.5±227.3、2 753.8±206.8;同时也高于单用rHBs免疫的小鼠。结论 CpG-ODN能增强接种rHBs产生的特异性淋巴细胞增殖反应。 Objective To explore the augment of CpG-ODN on cellular immune response induced by vaccination with recombinant HBV vaccine(rHBs).Methods BALB/c(H-2d) mice were inoculated intraperitoneally(i.p.)with rHBs at the doses ranging from 0.65-5.00 μg or CpG-ODN of 50.00 μg +rHBs at the doses ranging from 0.65-5.00 μg.4 weeks after the boosting immunization,spleen T lymphocytes were restimulated with rHBs in vitro,and specific helper lymphocyte proliferative activity was determined,the cpm for the spleen lymphocyte not being stimulated by rHBs were regarded as control.Results The cpm for specific helper lymphocyte proliferative activity in the group vaccinated with rHBs of 0.65,1.25,2.50,5.00 were 3 830.24±1 496.12,2 736.19±1 238.06,4 100.37±1 723.74,1 246.18±1 088.88,respectively,which were greater than those in control group with the spleen lymphocyte not being stimulated by rHBs:1 607.00±182.6,1 677.5±358.3,2 255.5±227.3,753.8±206.8(P〈0.05).The cpm for specific helper lymphocyte proliferative activity in the group vaccinated with rHBs+CpG-ODN was 6 863.87±1 243.24,6 582.48±1 132.48,7 332.43±938.28,5 184.24±947.45,which were greater than those in control group with the spleen lymphocyte not being stimulated by rHBs:2 307.00±182.6,2 577.5±358.3,2 655.5±227.3,2 753.8±206.8(P〈0.05),higher than those vaccinated with rHBs only.Conclusion CpG-ODN enhances the cellular immune response induced by vaccination with rHBs.
