摘要 目的:研究西洋参干预胰岛素抵抗大鼠活性部位的化学成分。方法:以乙醇提取,用大孔树脂吸附,乙醇洗脱得总皂苷,再用硅胶柱色谱及HPLC等分离技术与方法分离和纯化,通过理化常数及光谱数据确定其结构。结果:共分得3个化合物,经鉴定后分别证明为:6-O-[α-L-吡喃鼠李糖基-(1→2)-β-D-吡喃葡萄糖基]-20(24)-环氧-3,β6,α12,β25-四醇(Ⅰ);6-O-[α-L-吡喃鼠李糖基-(1→2)-β-D-吡喃葡萄糖基]-20-O-β-D吡喃葡萄糖基-达玛-24-烯-3,β6α,12,β20S-四醇(Ⅱ);(14S)-3-(4,5-二羟-6-(羟甲基)-3-(3,4,5-三羟基-6-(羟甲基)-顺-4-四氢吡喃-2-氧)-顺-4-四氢吡喃-2-氧)-4,4,8,10,14-五甲基-17-((S)-6-甲基-2-(3,4,5-三羟基-6-((3,4,5-三羟基-6-羟甲基)-顺-4-四氢吡喃-2-氧)甲基)-顺-4-四氢吡喃-2-氧)庚-5-烯-2-基)-2,3,4,5,6,7,8,9,10,11,14,15,16,17-十四氢-1H-环戊烯并[a]菲2-醋酸酯(Ⅲ)。 Objective:To study the chemical constituents of active site that American ginseng treats rat insulin resistance.Mefthods:They were extracted by ethanol and adsorbed by macro-reticular resins,then eluted with ethanol and isolated by silica gel column chromatography and HPLC preparation chromatography.Their structures were determined by physical and chemical properties,as well as spectrographical identification.Results:Three compounds were isolated from the active site that American ginseng treats rat insulin resistance.Their structures were identified as 6-O-α-L-rhamnopyranosyl-(1→2)-β-D-glucopyranosyl-dammar-20(24)-epoxy-3β,6α,12β,25-tetraol(Ⅰ).6-O-(α-L-rhamnopyranosyl-(1→2)-β-D-glucopyranosyl)-20-O-β-D-glucopyranosyl-dammar-24-ene-3β,6α,12β,20S-tetraol (Ⅱ).(14S)-3-(4,5-dihydroxy-6-(hydroxymethyl)-3-(3,4,5-trihydroxy-6-(hydroxy-methyl)-tetrahydro-2H-pyran-2-yloxy)-tetrahydro-2H-pyran-2-yloxy)-4,4,8,10,14-pentamethyl-17-((S)-6-methyl-2-(3,4,5-trihydroxy-6-((3,4,5-trihydroxy-6-(hydroxymethyl)-tetrahydro-2H-pyran-2-yloxy)methyl)-tetrahydro-2H-pyran-2-yloxy)hept-5-en-2-yl)-2,3,4,5,6,7,8,9,10,11,14,15,16,17-tetradecahydro-1H-cyclopenta[a]phenanthren-2-ylacetate(Ⅲ).
