碱性成纤维细胞生长因子对局灶性脑缺血再灌注大鼠海马组织中Notch1表达的影响 被引量:1

摘要 目的:检测Notch1在大鼠局灶性脑缺血再灌注海马组织的表达及碱性成纤维细胞生长因子(bFGF)的干预作用.探讨bFGF对局灶性脑缺血再灌注海马组织神经再生影响可能的分子调控机制.方法:采用大脑中动脉栓塞制作大鼠脑缺血再灌注模型.用免疫组织化学法和RT-PCR法检测海马组织Notch1的表达及bFGF的干预作用.结果:随着脑缺血再灌注,海马组织中Notch1 mRNA反应产物在第3天较假手术组增多,随缺血再灌注时间的延长逐渐增多,第7天表达最强,以后表达逐渐减少.Notch1 阳性细胞的变化趋势与Notch1 mRNA相似.注射bFGF后Notch1 的表达增加.结论:bFGF促进缺血再灌注后海马组织Notch1的表达,提示bFGF促进神经干细胞增殖作用可能由Notch信号通路介导. Objective: To investigate the effects of basic fibroblast growth factor (bFGF) on Notchl expression and hipp- oeampal neurogenesis in a rat model of cerebral ischemia/reperfusion, and to explore the related molecular mechanism. Methods: The model of transient focal ischemia was made by occlusion of the middle cerebral artery. The expression of Noteh1 and Notch1 mRNA in hippocampus was detected with immunohistochemical method and RT-PCR analyzed by microimage system. Results.. After three days following cerebral ischemia, Notehl mRNA in the hippoeampus were obviously in- creased. On the seventh day, Notch1 mRNA were more than any time. The expression of Notch1 reaction positive cells had similar trend with the expression of Notehl mRNA. Following bFGF injection, the expression of hippoeampal Notch1 had apparently increased in each group. Conclusion: bFGF promotes the expression of Notch1 in the hippocampus tissue. These findings indicate that bFGF can promote neural stem cell proliferation which may be mediated by Notch signaling pathway.
