摘要
目的探讨汉滩病毒A9株(HTNVA9)对肾实质细胞的致病作用。方法采用免疫标记技术和电镜技术观察HTNVA9株对人胚肾小球系膜细胞(MsC)的感染情况和病毒唑、磷甲酸钠对病毒感染的抑制作用。结果HTNVA9株能感染MsC并能在其中增殖,形成包涵体;病毒唑和磷甲酸钠能抑制HTNVA9株对MsC的感染,且加药时间愈早,抑制作用愈强。结论汉滩病毒感染是引起肾综合征出血热肾实质直接损伤的重要机制之一;为临床早期抗病毒药物治疗提供了实验依据。
Objective To study the pathogenesis of hantaan virus A 9 strain(HTNV A 9)to human mesangial cells(MsC).Methods The virus titer in culture fluid was measured by enzyme linked immunosorbent assay(ELISA).The HTNV antigen in MsC was detected by indirected immunofluoresence assay(IFA) and enzyme immunoassay(EIA).The shape and ultrastructure of infected MsC were observed by light microscope(LM) and transmission electron microscopy(TEM) respectively.Results HTNV infection to MsC was demonstrated by IFA,ELA and ELISA.By TEM,virion like grannules and viral inclusion bodies(IB) were observed to inhibit the replication of HTNV in MsC to some extent.The earlier the drugs like ribavirin and foscarnet used,the stronger the inhibitory effect was taken.Conclusion HTNV infection plays an important role in the pathogenesis of renal damage of Hemorrhagic Fever with Renal Syndrome.
