响应面法优化黄孢原毛平革菌产多糖培养基的研究

Optimization of fermentation conditions for polysaccharides of Phanerchaete chrysoprium based on response surface analysis

摘要 将Plackett-Burman设计法和响应面分析法相结合,对影响黄孢原毛平革菌(Phanerchaete chrysoprium)生产多糖的发酵培养基进行优化。结果表明,培养温度、酒石酸铵浓度和初始pH与多糖产量存在显著的相关性,采用优化后的条件进行摇瓶发酵,得到黄孢原毛平革菌产多糖的最佳工艺条件:葡萄糖10g/L,酒石酸铵0.4g/L,大量溶液60mL/L,微量元素液25mL/L,维生素B11mg/L,初始pH4.5,发酵温度37℃,接种量10%,非固定化培养5d。多糖产量从289.3mg/L提高到467.9mg/L。 Two level factorials designs of Plackett- Burman were constructed to select the Phanerchaete chrysoprium fermentation medium components of polysaccharides. Experimental results showed that culture temperature,ammonium tartrate concentration and initial pH all had significant influence on the polysaccharides production,and the optimal conditions were as follows:glucose 10g/L, ammonium tartrate 0.4g/L, the volume of mass element solution 60mL/L, the quantity of trace element solution 25mL/L, VB1 ling/L, initial pH4.5, culture temperature 37℃, inoculation quantity of 10%, nonimmobilized culture 5d. Under the optimal conditions, the polysaccharides production was increased from 289.3mg/L to 467.9mg/L.
