摘要
采用聚合酶链反应(PCR)方法改造鲑鱼(Oncorhynchustschawytscha)生长激素cDNA。改造后的cDNA去除了编码生长激素成熟肽cDNA以外的信号肽序列及5′端和3′端的非翻译序列,加入一起始密码,并在改造后的基因两端引入BamHI识别序列各1个。亚克隆后对改造过的基因进行了序列分析。
A polymerase chain reaction (PCR) method was used to modify the growth hormone cDNA of Oncorhynchus tschawytscha.The modified cDNA scquence contains an initiator codon and a completely coded mature peptide sequence of sGH for chinook trout,and deletes putative signal sequence and 5′ & 3 ′ untranstated regions.For easy to subclone,one Bam H Ⅰ site was added to each side of reformed gene.After cDNA subcloned,the modified gene sequcnce was analyzed.
