肝素化牛心包膜对VEGF_(165)控释的体外研究

摘要 通过交联的去细胞牛心包膜进行肝素化表面改性,提高对血管内皮细胞生长因子(VEGF)的控释能力及对内皮细胞(ECs)的促增殖能力。采用3种方式进行肝素改性,即共价键合肝素、离子结合肝素以及共价-离子联合负载肝素。评估肝素化基质对VEGF控释及促ECs粘附和增殖的影响。结果表明,共价键合肝素,结合VEGF稳定性强;而离子结合肝素,结合VEGF量可任意调节,且表面易于ECs粘附;共价-离子联合负载肝素,将两种方法的优点相结合,取长补短,使这种方式肝素改性的牛心包具有良好的VEGF控释能力和促ECs增殖能力。 This study is aimed to investigate the binding and release of vascular endothelial growth factor(VEGF) and its effects on adhesion and proliferation of endothelial cells(ECs) in acellular fresh specimens of bovine pericardiums,which were modified by heparinization.Crosslinked acellular fresh specimens of bovine pericardiums were heparinized by three motheds,including heparinized N-(3-dimethylaminopropyl)-N'-ethylcarbodiimide hydrochloride(EDC) treated acellular tissue samples,heparinized poly(ethyleneimine)(PEI) treated acellular tissue samples,and heparinized EDC-PEI treated acellular tissue samples.Controlled release of VEGF and its effects on adhesion and proliferation of ECs was evaluated.The binding of VEGF in heparinized EDC treated group was stable,while release of VEGF in heparinized PEI treated group was adjusted freely.Heparinized EDC-PEI treated group had the merit of combining the advantage of both EDC-Hep group and PEI-Hep group,showing beneficial effects on binding and release of VEGF and proliferation of ECs.
