甲基强的松龙对实验性变态反应性脑脊髓炎大鼠脑组织Nogo-A及其受体NgR蛋白表达的影响 被引量:2

摘要 目的探讨甲基强的松龙(MP)对实验性变态反应性脑脊髓炎(EAE)大鼠脑组织Nogo-A及其受体NgR蛋白表达的影响。方法将60大鼠随机分为正常对照组(n=10)和实验组(n=50)。实验组大鼠通过皮下注射粗制髓磷脂碱性蛋白建立EAE模型,造模成功大鼠随机分为EAE组(n=8)、大剂量甲基强的松龙(MP-H)治疗组(n=8)和小剂量甲基强的松龙(MP-L)治疗组(n=8)。大小剂量MP分别在模型成功后7d通过尾静脉注射,取脑前18h、6h各注射1次。各组标本均取视交叉前后2mm脑组织。HE染色观察脑组织的细胞形态学变化;免疫组化染色,检测Nogo-A及其受体NgR蛋白的阳性表达量。结果①HE染色:正常对照组未见异常;EAE组大鼠脑组织可见炎性细胞浸润、血管套、脱髓鞘改变及胶质细胞增生。②免疫组化染色:与正常对照组比较,EAE组Nogo-A及NgR蛋白阳性表达明显增加(P<0.05);与EAE组比较,MP-L组Nogo-A及NgR蛋白表达无明显变化,而MP-H组Nogo-A及NgR蛋白表达明显降低(P<0.05)。结论 EAE大鼠脑组织中Nogo-A及NgR蛋白表达增高;大剂量MP对EAE大鼠脑组织中Nogo-A及NgR蛋白的表达有一定的抑制作用,提示MP对EAE的作用机制可能与此作用有关。 ObjectiveTTo explore the expressions of Nogo-A and its receptor NgR protein in the brain of experimental allergic/autoimmune encephalomyelitis(EAE) rats treated with methylprednisolone(MP).MethodsSixty rats were randomly divided into two groups as normal control group(n =10) and treatment group(n =50) respectively.The EAE model was established by subcutaneous administration of coarse myelin basic protein(MBP).The EAE rats were randomly divided into control group(n =8) and high dose(30 mg/kg,MP-H) as well as low dose(10 mg/kg,MP-L) mythylprednisolone treated group(n =8 in each group) respectively.7 days later MP-H and MP-L rats were given mythylprednisolone 30 mg/kg and 10 mg/kg respectively via caudal vein.Rats were sacrificed at two time points(6 and 18 hr) after vein injection.Brain tissues 2 mm around optic chiasma were harvested and processed for further analysis.Morphological examination of rat brain tissues was done with HE staining under light microscope.The expressions of Nogo-A and its receptor NgR protein were detected by immunohistochemistry(IHC).Results① Under microscope with routine HE staining,no abnormality was found in the normal control group,and several features in EAE rats were observed,such as inflammatory cell infiltration,massive phagocyte encircling venula,demyelination,and gliocyte proliferation.② IHC Nogo-A and NgR positive cells in EAE group were significant more than those in normal rats(P〈0.05).Also,a statistically significant difference exits between MP-H and MP-L(P〈0.05).Two-two comparing,except for EAE group and MP-L group(P〈0.05),all others had statistical significance(P〈0.05).ConclusionsThe expressions of Nogo-A and Ng receptor in EAE group are increased.High dose of MP may depress the Nogo-A and NgR of the EAE rat brain tissue,and the mechanism of MP to EAE may be related to this inhibitory effect.
