摘要
以高效液相色谱法为分析手段,就溶液pH值、放置时间以及溶液稀释浓度对咪达唑仑共轭体系的影响进行了研究。结果表明,咪达唑仑在水中以开环物和闭环物的共轭体系形式存在;随着放置时间的延长,咪达唑仑共轭体系逐渐由开环物向闭环物转化,放置3 h后几乎完全转化为闭环物;溶液pH值增大时,开环物向闭环物转化,当pH值接近5时,几乎都转化为闭环物;反之溶液pH值降低时,闭环物向开环物转化,当pH<2.5时,开环物的含量随pH值的降低而快速上升;不同规格样品,稀释浓度越小,共轭体系中开环物的含量越高。
The effect of pH value of the solution,placing time and dilute concentration on the conjugated system of midazolam were studied by high performance liquid chromatography(HPLC) method.The result indicated that there was a conjugated system of open-ring form and closed-ring form of midazolam in water solution.The open-ring form of conjugated system of midazolam gradually transformed to closed-ring form with the increase of placing time and the conjugated system reached the dynamic equilibrium in 3 h.Meanwhile,the open-ring form of conjugated system of midazolam transformed to closed-ring form with the increase of pH value and there was almost only closed-ring form when pH5.On the contrary,the closed-ring form transformed to open-ring form with the decrease of pH value and the content of open-ring form rapidly increased when pH2.5.Moreover,the content of open-ring form increased with the decrease of dilute concentration of midazolam in different specifications.
