摘要
目的探讨镇肝熄风汤载药脑脊液对甲基-苯基-吡啶离子(1-methyl-4-phenylpyridinium,MPP+)诱导的PC12细胞c-Jun磷酸化的影响。方法镇肝熄风汤载药脑脊液预处理体外培养的PC12细胞后,加入MPP+诱导PC12细胞损伤模型。用Western blotting检测c-Jun和phospho-c-jun蛋白表达。结果 MPP+处理72h,与空白对照组比较,模型组phospho-c-jun蛋白表达显著上调,差异具有统计学意义(P<0.05)。与模型组比较,镇肝熄风汤载药脑脊液组phospho-c-jun蛋白表达显著下调,差异具有统计学意义(P<0.05)。而各组之间c-jun蛋白表达无显著性差异(P>0.05)。结论镇肝熄风汤载药脑脊液对MPP+诱导的PC12细胞c-Jun磷酸化具有抑制作用。
Objective To investigate the effect of cerebrospinal fluid containing Zhenganxifeng decoction（CSF-ZGXF） on c-Jun phosphorylation in PC12 cell induced by 1-methyl-4-phenylpyridinium ion（MPP＋）.Methods CSF-ZGXF were administrated 30 min before addition of MPP＋ into PC12 cell cutrure.Protein expression of c-Jun and phospho-c-jun was detected by western blotting.Results The protein expression of phospho-c-jun was up-regulate in MPP^＋ groups compared with the control group.That were downregulated in CSF-ZGXF group compared with the MPP^＋ group.Conclusion The results suggest that CSF-ZGXF suppress c-Jun phosphorylation in PC12 cell induced by MPP^＋.
