尼古丁对PDLFs细胞乙酰胆碱受体(nAChR)表达的影响

摘要 目的探讨尼古丁诱导的牙周膜成纤维细胞(periodontal ligament fibroblasts,PDLFs)凋亡的信号转导机制。方法本研究用1μg/ml,10μg/ml和100μg/ml不同浓度的尼古丁干预PDLFs细胞24h后,采用RT-PCR方法检测尼古丁乙酰胆碱受体(nAChR)的表达变化。结果不同浓度的尼古丁试验组(1μg/ml、10μg/ml和100μg/ml)作用于PDLFs细胞24h后,nAChR的mRNA表达水平显著增加,与正常对照组比较差异具有统计学意义(P<0.05),且随着尼古丁剂量的增加,nAChR的基因表达水平随之增高(r=0.948,F=254.231,P=0.000)。结论尼古丁致PDLFs细胞凋亡可能是通过nAChR起作用的。 Objective To study the signal transduction mechanism nicotine induced periodontal ligament fibroblasts(PDLFs)apoptosis.Methods This study used 1μg/ml,10μg/ml and 100μg/ml nicotine intervention PDLFs cells after 24h separately,and using RT-PCR to detect the expression levels of nicotine acetylcholine receptor(nAChR).Results Different concentrations of nicotine in the experimental group(1μg/ml,10μg/ml and 100μg/ml)acts on PDLFs cells for 24h,nAChR mRNA expression levels were significantly increased compared with the control group,and the difference is statistically significant(P〈0.05).nAChR gene expression increased with the increased dose of nicotine(r=0.948,F=254.231,P=0.000).Conclusion Nicotine induce PDLFs apoptosis may through the receptor of nAChR.
