摘要
目的:探讨补肾疏肝方对神经性厌食应激模型大鼠下丘脑-垂体-卵巢轴(hypothalamus-pituitary-ovarian axis,HPOA)功能的影响。方法:按照隔离、限食、束缚的方法建立神经性厌食大鼠应激模型,设立对照组、模型组、假操作组和补肾疏肝方组,每组10只,检测各组大鼠的体质量和动情周期的变化。在灌胃第35天、大鼠处于动情间期时收集血清和下丘脑、垂体组织标本,用放射免疫法检测血清雌二醇(estradiol,E2)、垂体卵泡刺激素(follicle stimulating hormone,FSH)、垂体黄体生成素(luteotrophic hormone,LH)和下丘脑β-内啡肽(β-endorphin,β-EP),用酶联免疫法测定血清皮质酮(corticosterone,CORT)。结果:补肾疏肝方组大鼠体质量明显高于假操作组(P<0.01);动情周期紊乱率明显低于模型组和假操作组;血清E2和垂体LH水平明显高于模型组(P<0.05);下丘脑β-EP明显低于假操作组(P<0.05);血清CORT水平明显低于模型组(P<0.05),亦明显低于假操作组(P<0.01);垂体FSH水平高于模型组和假操作组,但差异无统计学意义。结论:补肾疏肝方可以提高神经性厌食应激模型大鼠的血清E2和垂体LH水平,可能是通过降低下丘脑β-EP和血清CORT水平,进而起到调控HPOA功能的作用。
Objective:To investigate the effects of Bushen Shugan Recipe (BSSGR),a compound traditional Chinese herbal medicine,in regulating the hypothalamus-pituitary-ovarian axis (HPOA) in a rat model of stress-induced anorexia.Methods:Anorexia was induced in rats by the methods of separation,diet restriction and constraint.Rats were divided into 4 groups randomly:control group,untreated group,sham-operated group and BSSGR group.After the experiments,body weights and oestrous cycles of the 4 groups were compared.The levels of serum estradiol (E2),hypophysis luteotrophic hormone (LH),hypophysis follicle stimulating hormone (FSH) and hypothalamus β-endorphin (β-EP) were detected by radioimmunoassay.The level of serum corticosterone (CORT) was detected by enzyme-linked immunosorbent assay.Results:Body weight of BSSGR group was significantly increased in comparison with sham-operated group(P0.01); the oestrous cycle disordering rate was higher than those of the untreated group and sham-operated group; hypophysis LH and serum E2 were obviously increased in comparison with untreated group (P0.05); hypothalamus β-EP was obviously decreased in comparison with sham-operated group (P0.05); serum CORT was obviously decreased in comparison with untreated group (P0.05),and significantly decreased in comparison with sham-operated group (P0.01).Conclusion:BSSGR increased hypophysis LH and serum E2,and decreased serum CORT and hypothalamus β-EP in rats with stress-induced anorexia.
