摘要 目的观察藜芦主要成分原藜芦碱A对大鼠的心血管和呼吸系统的影响。方法实验分低、中、高和空白对照组共4组,每组10只,雌雄各半,给药组分别单次ig原藜芦碱A0.25,0.50和1.00 mg/kg,麻醉后行颈动脉插管术,观察原藜芦碱A对大鼠血压、ECG、呼吸频率及节律的影响。结果原藜芦碱A给药后可降低大鼠收缩压、舒张压、平均动脉压,减慢心率,延长QTc间期;可使大鼠呼吸频率减慢,呼吸幅度加大。上述变化均存在量效关系,给药150 min内均可逐渐恢复。结论原藜芦碱A对心血管和呼吸系统有一定的影响,能降低血压、减慢心率、延长QTc间期,能使呼吸频率减慢、呼吸幅度加大。 Objective To observe the effects of Protoveratrine A (PA) on cardiovascular and respiratory system in rats. Methods The experimental rats ( n = 40,m : f = 1 : 1 ) were equally randomized into 4 groups: blank control group and groups of low dose( ig , 0.25 mg/kg, single dose of PA), medium dose( ig, 0.50 mg/kg, single dose of PA) and high dose( ig, 1.00 mg/kg, single dose of PA). Carotid artery eannulation was established following anaesthesia in the rats and the effects of PA on the blood pressure, ECG, respiratory rate and width of rats were measured. Results The systolic blood pressure, diastolic blood pressure, mean arterial pressure, heart rate and respiratory rate of rats were decreased, the corrected QT (QTc) interval was prolonged and the respiratory width was increased in rats. The dose-effect relationships were observed for all the changes. However, these parameters returned to normal within 150 min. Conclusion PA has some effects on cardiovascular and respiratory systems in rats in reducing the blood pressure and heart rate, prolonging the QTc interval, decreasing the respiratory rate and increasing the respiratory width.
