摘要
目的探讨内毒素对肝硬化和正常大鼠肾脏功能的不同影响及其与肿瘤坏死因子α(TNFα)的关系。方法内毒素4mg/kg体重由尾静脉攻击四氯化碳合并乙醇所致肝硬化模型以及正常SD大鼠,分别于内毒素攻击后2、6、12小时检测肌酐(BUN)、尿素氮(CRE)和TNFα水平,并在光镜下观察肾脏组织学改变。结果肝硬化和正常大鼠在内毒素攻击后均出现肾功能不全,以6小时后较明显,但内毒素对肝硬化大鼠肾功能的损害更为显著,并且与TNFα水平呈正相关。光镜下二者均未见特异性器质性改变。结论肝硬化大鼠肾脏对内毒素损伤更为敏感,这种损伤主要是功能性改变,而TNFα是介导这种肾功能损害的重要介质之一。
Aim To study the different effect of endotoxin upon the renal function of cirrhotic and nomal rat and its relationship with tumor necrosis factor(TNF). Methods Endotoxin (4mg/kg body weight)was given to the cirrhotic rats induced by tetrachloride plus ethanol and normal SD rats through cauda vein.Blood urea nitroen,creatine and the level of plasma TNF were measured and the renal histological changes were observed by routine optical microscopy.Results Both cirrhotic and normal rats manifested renal insufficiency,especially 6 hours afer the endotoxin insult.The effect of endotoxin upon renal injury were much more significant for the cirrhotic rats and it was positively correlated with the plasma level of TNF.There was no specific changes for both the cirrhotic and normal rats under optical microscopy. Conclusion The kidney of cirrhotic rat was much more sensitive to the injury of endotoxin and TNF was one of the important mediator for this kind of functional changes.
