摘要
目的:探讨外源一氧化氮(nitric oxide,NO)对胃癌AGS细胞生长及c-jun和c-fos表达的影响。方法:应用MTT法评价NO供体硝普钠(sodium nitroprusside,SNP),SNP的同源类似物铁氰化钠Na_3Fe(CN)_6和一氧化氮合酶(nitric oxide synthase,NOS)抑制剂N-硝基-L-精氨酸甲酯(N-nitro-L-arginine methylester,L-NAME)对胃癌细胞生长的抑制作用,RT-PCR和免疫印迹检测c-jun和c-fos的基因和蛋白表达。结果:SNP剂量依赖性的抑制AGS细胞的生长,其IC_(50)值为2.13mmol/L。1mmol/L SNP处理12、24、48h后,细胞生长抑制率分别为(16.12±0.85)%、(20.33±1.31)%、(22.06±2.03)%(P<0.05),NOS抑制剂L-NAME预处理明显减弱了SNP对癌细胞的生长抑制效应,Na_3Fe(CN)_6对胃癌细胞生长无抑制。与对照组比较,SNP使c-jun和c-fos的mRNA和蛋白表达降低,NOS抑制剂L-NAME预处理逆转了SNP的这种作用,Na_3Fe(CN)_6对c-jun和c-fos的mRNA和蛋白表达无影响。结论:NO可诱导胃癌细胞c-jun和c-fos表达降低,抑制癌细胞生长。
Objective: To investigate the effect of exogenous nitric oxide on the growth and the expressions of c-jun and c-fos in gastric cancer cell line AGS. Methods: The AGS cells were treated with SNP, Na3Fe(CN)6 and L-NAME. The growth inhibition of cancer cells was determined by MTT assay. The changes of mRNA expressions of c-jun and c-fos were examined by RT-PCR. The protein expressions of c-jun and c-fos were analyzed by Western blot. Results: SNP inhibited AGS cell growth in a dose-dependent manner with a 24 hour IC50 value of 2.13 mmol/L. When the AGS cells were treated with 1 mmol/L SNP for 12, 24 and 48 hours, the growth inhibition rates were (16.12 ±0.85)%, (20.33±1.31)% and (22.06 ± 2.03)%, respectively (P〈0.05). L-NAME reversed the effect of SNP in inhibiting the growth of AGS cells. Na3Fe(CN)6 had a very small effect on the growth inhibition of AGS cells. Compared with the control group, SNP reduced the gene and protein expressions of c-jun and c-fos in gastric cancer cell line AGS. Such decreases were reversed by pretreatment with L-NAME. Na3Fe(CN)6 had little effect on the gene and protein expressions of c-fos and c-jun. Conclusion: Nitric oxide may work to inhibit gastric cancer cell line AGS growth by decreasing the expression of c-fos and c-jun.
