摘要
目的观察本研究所研制的重组人色素上皮细胞衍生因子(rhPEDF)对大鼠视神经夹挫伤后的保护作用。方法健康SD大鼠32只,随机分为4组:对照组及rhPEDF不同浓度(1μg/μL、0.1μg/μL、0.01μg/μL)的3个组。均制成左眼视神经夹挫伤模型,分别于损伤前,损伤后即刻,1周后记录左眼闪光视觉诱发电位(F-VEP)。于损伤当天,第2,4,6天向左眼球后注射各组液体5μL。用药1周后,摘取眼球及视神经,HE染色,光镜下观察形态学改变。结果损伤后,各组F-VEP波峰潜时(P1)均延长,波幅均降低(N1-P1),与损伤前相比差异有统计学意义(P<0.05),造模成功。用药7d后,各组P1值恢复至损伤前,波幅(N1-P1)值有一定恢复。rhPEDF高浓度组与其它3组相比,差异有统计学意义(P<0.05),但仍未恢复至损伤前水平(P<0.05)。结论本研究所重组的rhPEDF具有神经营养功效。为进一步开发一种新的眼部神经营养因子奠定基础。
Objective To investigate the protection effect of recombinant human epithelium derived factor(rhPEDF) produced by our institute on rats after optic nerve injury.Methods Thirty two healthy SD rats were randomly divided into 4 groups:PBS group and rhPEDF(1 μg/μL,0.1 μg/μL,0.01 μg/μL) groups.Optic injury was made on the left eye in the above groups.The left eye flash-visual evoked potential(F-VEP) was recorded at pre-injury,immediate after injury and 7days after injury.Five microliter different liquid was injected into retrobulbar in each group on the same day,the first 2,4,6 days after injury.After 7d medication,the eye and optic nerve were removed,stained by HE and then the morphological changes were observed under light microscope.Results The latency of P1 wave(P1) was delayed and the amplitude of P1 wave(N1 -P1) was immediately reduced in each group after injury.Compared with the results of pre-injury,the data were statistically significant(P〈0.05) and the model were successfully made.After 7d medication,P1 values of each group returned to pre-injury levels and the amplitude of P1 wave(N1-P1) restored a certain value.Compared with the other three group,the data of high concentration of rhPEDF group were statistically significant(P〈0.05),but did not recovered to pre-injury levels(P〈0.05).Conclusions The rhPEDF that made by our institute shows neurotrophic effects on injured optic nerves and this work lay the foundation for further the development of a new neurotrophic factor.
