雷丸药材定性和定量分析方法研究 被引量:7

摘要 目的:建立雷丸主要成分的定性与定量分析方法。方法:采用薄层色谱法对雷丸中的麦角甾醇、麦角甾酮进行鉴别。运用可见-紫外分光光度法对雷丸所含的驱绦虫成分雷丸凝集素进行含量测定,以水为提取溶剂,采用Fonlin酚法,测定波长650nm。结果:应用所建立的方法对13批雷丸药材进行定性与定量分析,结果TLC斑点清晰,专属性强,均可检出雷丸的特征斑点;13批雷丸药材雷丸凝集素的含量以牛血清白蛋白计为0.61%~4.23%;方法学研究结果表明,牛血清白蛋白的量在52.76~263.8μg范围内呈良好的线性关系(r=0.9997),平均回收率为98.0%(RSD=1.0%,n=6)。结论:本方法操作简便,重复性好,结果准确可靠,可用于雷丸的定性定量分析,以控制雷丸药材及相关产品的质量。 Objective:To establish the qualitative and quantitative analysis methods of Omphalia.Methods:With TLC to identify ergosterol and ergosta-4,6,8(14),22-tetraen-3-one.Ultraviolet spectrophotometry and colourmetry were used to determinate Omphalia lapidescens lectin(OLL) of Omphalia by Folin phenol agent,and the detected wavelength was 650 nm.The extracting solvent was water.Results:In TLC the spots developed were quite clear and exclusive.The contents of OLL in 13 lots of samples were 0.61%-4.23%,the linear range of senum albumin bovine was 52.76-263.8 μg(r=0.9997),the average recovery was 98.0%(RSD=1.0%,n=6).Conclusion:The method is simple,accurate and can be used for the qualitative and quantitative analysis of Omphalia and also related products.
