摘要
目的:建立雷丸主要成分的定性与定量分析方法。方法:采用薄层色谱法对雷丸中的麦角甾醇、麦角甾酮进行鉴别。运用可见-紫外分光光度法对雷丸所含的驱绦虫成分雷丸凝集素进行含量测定,以水为提取溶剂,采用Fonlin酚法,测定波长650nm。结果:应用所建立的方法对13批雷丸药材进行定性与定量分析,结果TLC斑点清晰,专属性强,均可检出雷丸的特征斑点;13批雷丸药材雷丸凝集素的含量以牛血清白蛋白计为0.61%～4.23%;方法学研究结果表明,牛血清白蛋白的量在52.76～263.8μg范围内呈良好的线性关系(r=0.9997),平均回收率为98.0%(RSD=1.0%,n=6)。结论:本方法操作简便,重复性好,结果准确可靠,可用于雷丸的定性定量分析,以控制雷丸药材及相关产品的质量。
Objective：To establish the qualitative and quantitative analysis methods of Omphalia.Methods：With TLC to identify ergosterol and ergosta-4,6,8（14）,22-tetraen-3-one.Ultraviolet spectrophotometry and colourmetry were used to determinate Omphalia lapidescens lectin（OLL） of Omphalia by Folin phenol agent,and the detected wavelength was 650 nm.The extracting solvent was water.Results：In TLC the spots developed were quite clear and exclusive.The contents of OLL in 13 lots of samples were 0.61%-4.23%,the linear range of senum albumin bovine was 52.76-263.8 μg（r=0.9997）,the average recovery was 98.0%（RSD=1.0%,n=6）.Conclusion：The method is simple,accurate and can be used for the qualitative and quantitative analysis of Omphalia and also related products.
