柯萨奇病毒B_1接种豚鼠建立多发性肌炎模型 被引量:3

Study on coxsackievirus B1induced polymyositis in guinea pig models

摘要 目的探讨柯萨奇病毒B1(CVB1)感染与多发性肌炎(PM)发病的关系,利用CVB1接种豚鼠建立PM模型。方法分别用0.1ml(A组)和0.3ml(B组)毒力(TCID50,即50%细胞感染剂量)为10-5的CVB1分别一次性接种24只和36只豚鼠,并用3mg(阈下剂量)兔肌匀浆加完全弗氏佐剂每周免疫1次;另有10只豚鼠,仅用3mg兔肌匀浆加完全弗氏佐剂每周免疫1次为对照组(C组),共5周。结果A、B组3周后均有豚鼠出现四肢肌无力等症状,肌酶谱明显升高,豚鼠股肌病理呈局灶性炎性改变,C组无异常。A、B组5周后的肌酶谱升高阳性率存在显著差异(A组为11.8%,B组为38.2%)。结论CVB1感染是PM发病诱因之一,其毒力及接种量与PM的发病相关。 Objective To study the relation between infection with coxackievirus B1(CVB1) and onset of polymysitis in the polymyositis models of guinea pigs infected with CVB1. Method Sixty guinea pigs were innoculated with 0.1 ml of CVB1 in group A and with 0.3 ml of CVB1 in group B. The CVB1 toxicity being 10-5 TCID50. Then they were immunized once a week with 3 mg rabbit muscle homogenates in Freunds complete adjuvant, ten guinea pigs immunized once a week only with 3 mg rabbit muscle homogenates in Freunds complete adjuvant as a control (group C). Results Some guinea pigs innoculated with CVB1 showed muscular weakness of their limbs, the serum enzymes were evidently elevated and the histopathological, muscular changes of polymyositis were confirmed 3 weeks after the viral infection, the guinea pigs of group C were normal. The positive rates of elevation in muscle enzymes were significantly different between the two groups of guinea pigs. In group A elevation in muscle enzymes was found in 11.8%, whereas in group B it appeared in 38.2% five weeks after the viral infection. Conclusion Not only infection with coxsackievirus B1 could induce PM but also its quantity and toxicity were correlated with the onset of polymyositis.
