摘要
目的:考察垂盆草总黄酮中已确证的8种单体成分对肝细胞的保护作用,为垂盆草药材质量控制指标成分的筛选提供依据。方法:取已经确证的垂盆草总黄酮中的8种单体成分,用二甲基亚砜(DMSO)溶解,临用前用培养液稀释成5个不同的浓度,分别用MTT法检测其对H2O2诱导的人系L02肝细胞损伤的保护作用。结果:模型组OD值与正常组比较差异有极显著性,金丝桃苷(10-1~10-3g·L-1)、5,4′-二羟基-8,3′-二甲氧基黄酮-3,7-二-O-β-D-葡萄糖苷(1.0~10-2g·L-1)与模型组比较差异有极显著性,而金丝桃苷(10-4g·L-1)、苜蓿苷(1.0~10-1g·L-1)和槲皮素(1.0~10-2g·L-1)与模型组比较差异有显著性。结论:金丝桃苷、5,4′-二羟基-8,3′-二甲氧基黄酮-3,7-二-O-β-D-葡萄糖苷在一定浓度范围内对H2O2所致L02肝细胞损伤有显著的保护作用,苜蓿苷和槲皮素也有一定的保护作用。
OBJECTIVE To observe the protective effects of the eight confirmed flavonoids compositions from Hebra Sedi Sarmentosi on liver cells, to provide basis for selecting the index composition of quality control for Hebra Sedi Sarznentosi. METHODS Eight confirmed flaconoids canpositions were taken, dissolved by DMSO, diluted to five different concentrations by culture solution before used, then used the method of MTT to check the protective effect on L02 cell injured by H2O2. RESULTS Compared with the normal group,the OD level of model group has extremely significant difference,compared with the Model group, hypericin ( 10^-1~10^-3g·L^-1 ), 5,4'-dihydroxy-8', 3'-dimethoxy-3,7-flavonoids-2-O-β-D-glucosid ( 1.0-10^-2g·L^-1) had extremely significant difference, hypericin( 10^-4g·L^-1 ), tricin-7-O-β-D-lucopyranoside( 1.0-10^-1g·L^-1 ) and quercetin( 1.0- 10^-2g·L^-1 ) had significant difference. CONCLUSION Hyperiein and 5,4'-dihydroxy-8', 3'-dimethoxy-3,7-flavonoids-2-O-β-D- glucosaide have protective effect on liver cell in vitro injured by H2 02, quercetin and tricin-7-O-β-D-lucopyranoside also have some effects on certain degree.
