醋柳黄酮对大鼠心肌缺血-再灌注损伤及MMP-9表达的影响

摘要 目的观察醋柳黄酮(TFH)对大鼠心肌缺血-再灌注后损伤及组织MMP-9表达的影响,探讨对心肌缺血-再灌注损伤后保护机制。方法将50只大鼠随机分组:模型组、氯沙坦组、醋柳黄酮大、小剂量组及假手术组。结扎左冠状动脉前降支30min后松开制作心肌缺血-再灌注模型。对心肌组织进行HE染色观察心肌坏死的面积及病理变化,免疫组织化学检测再灌注心肌组织MMP-9表达。结果 HE切片观察TFH大、小剂量组预处理的大鼠心肌坏死较模型组明显减轻,免疫组化结果显示TFH大、小剂量组较模型组MMP-9表达减少,有统计学意义。结论TFH对再灌注损伤心肌有保护作用,其作用机制与抑制心肌中MMP-9表达有关。 Objective To observe the effects of total flavones of hippophae rhamnoides L on myocardial ischemia-reperfusion(I-R)injury and the MMP-9 expression in cardiac tissues of hypertensive rats,and to explore the protective mechanism.Methods 50 rats with hypertension were randomly divided into model,losartan,different dosages of sindacon and sham groups.Myocardial I/R injury model was established by ligating left anterior descending coronary artery for 30 minutes and then releasing.The cardiac muscle tissue was stained by HE to observe its necrosis area and pathological changes as well as the expression of MMP-9 by immunohistochemistry.Results Necrosis of cardiac muscle in hypertensive rats was significantly relieved in different dosages of sindacon groups compared with that of model group,and the expressions of MMP-9 of different dosages of sindacon groups were decreased compared with that of model group.Conclusions Sindacon has protective effect on cardiac muscle after myocardial I/R injury,and its mechanism maybe related to its inhibition of the MMP-9 expression in cardiac muscle.
