摘要
目的在视觉发育可塑性关键期终止前后,观察大鼠视皮层内NR2A/NR2B比率的发育性变化。方法健康SPF级LongEvans大鼠15只,雌雄不限,采用Western blot法观察出生后3、4、5、6、7周正常大鼠视皮层中NR2A和NR2B的蛋白表达量,分析NR2A/NR2B随着大鼠视皮层发育表达的变化。结果 Western blot检测显示,生后3~7周正常大鼠视皮层中NR2A蛋白的表达量逐渐增加,差异有统计学意义(F=156.263,P=0.008),生后4、5、6、7周组大鼠视皮层中NR2A的积分光密度(IOD)值明显高于生后3周组大鼠,差异均有统计学意义(P<0.05)。可塑性关键期终止前后,正常大鼠视皮层中NR2B蛋白的表达量逐渐降低(F=73.340,P=0.007),生后6~7周趋于稳定。可塑性关键期终止前后,大鼠视皮层中NR2A/NR2B比率随着大鼠视皮层的发育逐渐增加,差异有统计学意义(F=113.031,P=0.003)。结论在可塑性关键期终止前后,NR2A/NR2B比率的变化参与了视皮层可塑性终止的过程。
Background During a critical period of early postnatal development,visual system changes in both shape and function to adapt to the regulation of external environment and internal gene.N-mehyl-D-aspartate (NMDA) receptor plays an important role in the visual cortical plasticity,and the subunit 2A of NMDA receptor (NR2A) and NR2B are the master subunits of NMDA receptor.However,the ratio of NR2A/NR2B in rat visual cortex at terminal stage of critical period is unclear.Objective The aim of this study was to investigate the changes of NR2A/NR2B in rat visual cortex at terminal stage of critical period of visual plasticity.Methods Normal SPF Long Evans rats aged postnatal 3 weeks,4 weeks,5 weeks,6 weeks,7 weeks were collected and 3 rats for each.Expression of NR2A and NR2B proteins in normal rat visual cortex with different age were detected using Western blot assay.The use of the rats followed the Statement of Use of Experimental Animal by Third Military Medical University.Results Western blots results showed that the level of NR2A in normal rat visual cortex was gradually increased from postnatal 3 weeks group through postnatal 7 weeks group (F=156.263,P=0.008).The average integral average optical densit values (IOD) of NR2A in postnatal 4 weeks group,postnatal 5 weeks group,postnatal 6 weeks group,postnatal 7 weeks group were higher than that of postnatal 3 weeks group (P0.05).The expressive level of NR2B in normal rat visual cortex was gradually declined from postnatal 3 weeks group through postnatal 7 weeks group(F=73.340,P=0.007).The NR2A/NR2B ratio in normal rat visual cortex was significantly elevated with age (F=113.031,P=0.003).Conclusion The changes of NR2A/NR2B ratio is involved in the terminal process of visual plasticity period.
