皂角刺总黄酮对HCT116细胞增殖和凋亡的影响 被引量:5

Effects of flavone components from Spina gleditsiae on proliferation and apoptosis of HCT116 cell line

摘要 目的:研究皂角刺总黄酮对HCT116细胞增殖和凋亡的影响,探讨其抗肿瘤作用机制;方法:体外培养人结肠癌HCT116细胞,用不同浓度的皂角刺总黄酮(12.5、25、50、100、200和400mg/L)处理后,采用CCK-8试剂盒(cell countingkit-8)检测对HCT116细胞增殖的抑制作用,Hoechst33258荧光染色观察细胞凋亡的形态学变化,流式细胞仪Annexin V/PI双染法检测细胞的凋亡率。结果:不同浓度的皂角刺总黄酮作用于细胞48h后,能显著抑制HCT116细胞的增殖,随着药物浓度的增加,抑制作用增强,IC50为104.72±0.96mg/L;Hoechst33258染色可见细胞核固缩、碎裂和凋亡小体等凋亡形态学改变;流式Annexin V/PI双染法检测显示,不同浓度的皂角刺总黄酮均可诱导HCT116细胞凋亡,在100mg/L时细胞凋亡率为(35.61±3.76)%。结论:皂角刺总黄酮能明显抑制HCT116细胞的增殖和诱导其凋亡。 Objective:To investigate effects of flavone components from Spina gleditsiae on proliferation and apoptosis of HCT116 cell line,and explore its anti-tumor mechanism action.Methods:Human colon cancer HCT116 cells were cultured with RPMI-1640 medium,and treat HCT116 cells with different concentrations(12.5,25,50,100,200) mg/L of FSG after 24,48 and 72 hours'.The inhibition of cell proliferation was determined by CCK-8(Cell Counting Kit-8) assay.The morphological changes of apoptosis was observed by Hoechst33258 staining.The apoptosis rate of HCT116 cells was observed by Flow Cytometry with Annexin-V/PI staining.Results:The flavone components from Spina gleditsiae significantly inhibited the proliferation of HCT116 cells after 48 hour',and which in a dose-dependent manner,the IC50 value was 104.72 0.96 mg/L.After treatment of different concentration of flavone components from Spina gleditsiae,typical morphological changes of cell apoptosis such as karyopyknotic pyknic hyperfluorescence bolus,nuclear fragmentation and apoptotic body were observed by Hoechst 33258 staining.The apoptosis of HCT116 cells was induced by different concentration of flavone components from Spina gleditsiae,and the apoptosis rate was(35.00 ± 3.76) % at 200 mg/L by using Flow Cytometry with Annexin-V/PI staining.Conclusion:Flavone components from Spina gleditsiae can inhibit the proliferation of HCT116 cells and induce their apoptosis.
