摘要
目的:采用淫羊藿苷icarrin(ICA,从朝鲜淫羊藿中提取的中药单体),作用于人急性早幼粒白血病细胞HL-60,较系统地对ICA诱导肿瘤细胞凋亡进行了观察,为ICA的开发应用提供实验依据.方法:采用电镜、流式细胞术、DNA片段化检测、RT-PCR等技术进行研究.结果:ICA在体外诱导HL-60细胞凋亡,呈典型的细胞凋亡形态学和生化特征,并具有时间剂量依赖性.ICA诱导肿瘤细胞凋亡的同时,下调相关基因bcl-2和c-myc基因mRNA和蛋白表达水平.结论:ICA体外诱导HL-60细胞凋亡,其机理可能与下调bcl-2和c-myc基因mRNA和蛋白表达水平密切相关.
To examine if human acute promyelocytic leukemia cells (HL-60) apoptosis of could be inducedby icarrin(ICA, a mononer purified from Epimedium Koreanum Nakai). Methods: The cells undergoing apoptosis were determined by morphology, DNA gel electrophoresis and flow cytometry (FCM). The expression of apoptosis associated genes bcl-2 and c-myc were evaluated by FCM and semi-quantitative RT-PCR methods. Results: ICA induced apoptosis showed classical morphological and biochemistry features in human cancer cells in a time and dose dependent manner in vitro. While inducing apoptosis, ICA downregulated expression of apoptosis associated genes bcl-2 and c-myc. Conclusion : This study suggests that ICA induced apoptosis of HL-60 cells is associated with downregulation of bcl-2 and c-myc genes rnRNA and protein expression.
