骨膦抗去坐骨神经雄鼠骨质疏松实验研究 被引量:1

Experimental study on anti-osteoporosis effect of clodronate in sciatically denervated male rats

摘要 对8月龄雄性大鼠切断坐骨神经后皮下注射骨膦8周(BP组),与单纯去坐骨神经组(SN组)及假手术组(SHAM组)作比较,采用骨生物力学、骨病理组织学及骨形态计量学等方法对骨膦的药效进行评价。结果:与SHAM组比较,SN组骨抗弯力、骨小梁体积密度及骨小梁平均宽度、成骨细胞指数明显降低,而破骨细胞指数及尿羟脯氨酸升高;BP组各项指标则与SHAM组无显著性差异。提示:骨膦可有效防止坐骨神经切断所致的失骨。 To evaluate the anti-osteoporosis effect of clodronate in sciatically denervated male rats, 24 male Wistar rats, aged 8 months, were divided randomly into 3 groups: a) the simple sciatically denervated (SN) group which were subjected to sciatic nerve resection in left tibia; b) the clodronate (BP) group which were subjected to sciatic nerve resection in left tibia and treated on the next day with clodronate 5mg/kg BW by subcutaneous injection twice a week for 8 weeks; and c)the sham-operated (SHAM) group. All the rats were sacrified after the operation. The blood and urine samples were collected for determination of biochemical indices. The bumeri and tibiae were prepared for determination of bone biodynamics, histopathology and histomorphometry. The results showed that as compared with the SHAM group, the bending resistance of bones, the trabecular bone volume and thickness as well as the osteoblast index were all significantly reduced in the SN group, while its osteoclast index and urine hydroxyproline content were obviously increased. In the BP group, however, these indices were close tonormal levels. The data suggested that clodronate can effectively prevent bone loss in sciatically denervated male rats.
