摘要
目的:观察中药清化散对创伤性异位骨化模型大鼠骨形态发生蛋白-4(BMP-4)的影响。方法:80只SD大鼠,除16只作为正常对照组外,其余64只行右侧跟腱切断术,建立创伤性异位骨化大鼠模型,造模后第18天,除正常对照组外,将造模后大鼠随机分为4组,分别是模型组、局部外用清化散治疗组、口服西乐葆治疗组、局部外用清化散并口服西乐葆治疗组,分别给予相应的药物治疗。用药32、61天后每组各处死一半大鼠,取组织标本,检测跟腱组织内BMP-4的含量。结果:用药32天后,模型组的BMP-4含量明显高于正常对照组(P<0.01)和治疗组(P<0.05)。各治疗组的BMP-4含量均较对照组稍高,但无显著性差异(P>0.05),各治疗组之间也无显著性差异(P>0.05)。用药61天后,模型组的BMP-4含量较30天前增幅更大,明显高于正常对照组(P<0.01)和治疗组(P<0.05)。各治疗组的BMP-4含量较30天前降低,接近正常对照组,与对照组之间无显著性差异(P>0.05)。结论:中药清化散、西乐葆或者二者联合应用均能抑制跟腱组织内BMP-4的合成,从而抑制创伤性异位骨化的发生。
Objective:To observe the effects of Chinese herb Qinghuasan on Bone morphogenetic protein-4(BMP-4)in Sprague-Dawley(SD)rat of traumatic heterotopic ossification.Method:80 SD rats were observed,16 SD rats were observed as normal control group,64 SD rats were performed right Achilles tenotomy to induce traumatic heterotypic ossification and 18 days later they were divided into 4 groups randomly(n=16):model group,group treated with local administration of Qinghuasan,group treated with oral administration of celebrex,group treated with both local administration of Qinghuasan and oral administration of caliber.Half rats of each groups were executed respectively after 32 and 61 days' treatment,the content of BMP-4 of the Achilles tendon were detected.Result:After 32 days' treatment,the content of BMP-4 in model group was significantly higher than the normal control group(P〈0.01) and treated groups(P〈0.05).The content of BMP-4 in each treated group was higher than the one in control group,but there was no significant difference between them(P〈0.05),and no significant difference between each two treated groups(P〈0.05).After 61 days' treatment,the content of BMP-4 in model group was significantly higher than the one in normal control group(P〈0.01)and treated groups(P〈0.05),which was much higher than 30 days before.The content of BMP-4 in each treated group was lower than 30 days before and close to the content in normal control group,there was no significant difference between treated groups and control group(P〈0.05).Conclusion:Qinghuasan,Celebrex,or combination of them can all inhibit the synthesis of BMP-4 in Achilles tendon,and then inhibit the occurrence of traumatic heterotopic ossification.
