摘要
在优化的固定化条件下,通过戊二醛交联直接将青霉素酰化酶固定化。在优化的环境条件下测定游离酶和两种固定化酶的动力学常数。结果表明,尽管固定化酶的米氏常数增大,但产物抑制作用减弱,裂解青霉素的实验结果表明,固定化酶更适合在工业上应用。
Penicillin acylase was directly immobilized with glutaraldehyde as the crosslinked agent under the optimal immobilized conditions.The dynamic constans of free enzyme and two kinds of immobilized enzymes were determined under the optimal environmental factors.The results showed that Km of immobilized enzyme was obviouslt increased,but the inhibitions of products were decreased.The experiments of hydrolized penicillin-G showed that immobilized enzyme was suitable for utilization on industrial scale..
